The unbeaten Brough, who is rebuilding his career after a serious hand injury, joins fellow scousers Derry Mathews, Stephen Jennings and Terry Needham at Olympia.
Farrell claimed the Central Area title in June with a fifth round stoppage of Joe Elfidh and he’ll want to upset the hometown heroes along with Dewsbury’s former British super featherweight champion Gary Sykes and Halifax man Liam Patrick Walsh.
Just one spot remains available for the 26th edition of the eight-man tournament, which co-features Merseysider Rocky Fielding in action against Wayne Reed.
“The line up for Rocky and Betfair Prizefighters just gets better and better,” said promoter Eddie Hearn. “Nathan is a great local unbeaten talent and I’ve been really impressed with Kieran, who recently boxed on our Carl Froch show in May.
“The event will be an immediate sell out and we have around a dozen fighters vying for the final spot.”
Fielding will challenge Reed for the Central Area super middleweight belt in his first title fight at the venue of his record breaking Prizefighter triumph in March 2011.
Reed won the title earlier this month with a fifth round stoppage of Yorkshire’s Alistair Warren at Hillsborough.
